Anticancer and apoptotic effect of compounds from medicinal Plants on lung cancer cells

Dr. Biba Vikas carried out her Post Doctoral degree in Applied Sciences &Technology- Biotechnology University of Kerala, India. Later, She started working as a Lecturer In PMSA, Kerala, India. Presently, she is working as a post doctoral fellow in Regional Cancer Center, JNTBGRI, Kerala, India.

Natural products represent rich and largely untapped reservoirs of structural and chemical diversity for the discovery of drugs with potent applications in the treatment of diseases. Annona squamosa (Annonaceae), possess wide range of biologic activities. Simarouba glauca in Simaroubaceae family has a long history in herbal medicine in many countries. Present study was undertaken to evaluate the cytotoxicity and apoptotic potential of the active principles isolated from Simarouba glauca leaves and Annona squamosa seeds. Evaluated cell based assays and validated in murine model. Compounds isolated from these plants were purified and chemically characterized IMD and SGAF. Cytotoxicity studies revealed cell death in dose and time dependent manner. Fluorescence microscopic analysis using Acridine orange-Ethidium bromide dual stain and Hoechst-33342 stain revealed apoptotic nature of compound induced cell death at 5 µg/ml. Flowcytometric analysis revealed a higher percentage of AnnexinV positive cells by the treatment with compounds, compared to untreated control cells, indicating induction of early apoptosis. Caspase-3 activation of the treated cells suggests the possible caspase mediated mechanism of apoptotic induction. There was significant tumor reduction in mice model. Suggesting these compounds to be further validates for potential application as anticancer drugs.
